SUMMARY:

“Cell culture is an essential technique in basic research, but it still faces limitations in methodological quality and transparency in reporting, which compromises the reproducibility and validity of scientific findings. Although there are methodological guidelines for other types of research, there remains a specific gap for in vitro studies involving cell culture. To address this gap, this thesis proposes the development and validation of PETRICCS (Preferred and Transparent Reporting Items for Cell Culture Studies), a methodological guide designed to assist in the preparation and reporting of studies with improved methodological quality. The thesis is structured in four stages: (1) development of guidelines in the form of a checklist, flowchart, and step-by-step instructions; (2) practical validation of clarity, applicability, and usability by independent experts; (3) adaptation of PETRICCS as a critical appraisal tool for assessing risk of bias in systematic reviews; and (4) elaboration of an educational project, including the development of a guiding e-book. Partial results of the first stage include the development of PETRICCS essential items, created in collaboration with a multidisciplinary steering committee, and the implementation of a pilot study — a modified Delphi process conducted between September 2024 and April 2025, with the participation of 17 graduate students experienced in cell culture. The identified items were organized into seven main thematic domains and were rated for relevance by the pilot study participants. This phase allowed for the initial refinement of the tool prior to the completion of stage 1 and progression to stage 2. In stage 3, a critical appraisal tool was developed based on PETRICCS essential items, adapted into question format with standardized answers ("Yes", "No", "Unclear"), inspired by consolidated models such as SYRCLE, OHAT, and CONSORT. This version was applied in a systematic review in partnership with a collaborating laboratory, which allowed for assessment of its applicability and the implementation of necessary adjustments for the completion of stage 3. Overall, the PETRICCS guideline aims to strengthen transparency, reproducibility, and methodological quality in cell culture studies, from experimental planning to the final manuscript writing. The next phases of the study include a new Delphi round with international experts, conducted via the eDelphi.org platform, for the refinement (completion of stage 1) and validation of the final checklist (stage 2). Additionally, a scientific article and an educational e-book will be prepared to broadly and practically disseminate PETRICCS within the scientific community (stage 4).”
